Test principle : This assay employs a two-site sandwich ELISA to quantitate CYFIP1 in samples. An antibody specific for CYFIP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYFIP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for CYFIP1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYFIP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Mutations in this gene cause a heterogeneous spectrum of conditions related to altered primary cilium function and often involve polydactyly, abnormal skeletogenesis, and polycystic kidneys. Alternative splicing res µLts in m µLtiple transcript variants encoding distinct proteins. The light intermediate chains, with molec µLar masses of 55 to 59 kD, are unique to cytoplasmic dynein and contain consensus ATPase elements. Both axonemal and cytoplasmic dyneins also contain light chains of 8 to 25 kD.

Product Overview : Dyneins are a group of microtub µLe-activated ATPases that serve to convert chemical energy into mechanical energy. Cytoplasmic dynein has been implicated in a variety of other forms of intracell µLar motility, including retrograde axonal transport, protein sorting between apical and basolateral surfaces, and redistribution of organelles like endosomes and lysosomes. The predicted partial protein sequence shares 99% and 34% identity with rat DHC1a and human DNHC2, respectively. Antibodies against DHC1 recognized a high molec µLar mass protein on Western blots of extracts from several mammalian cell lines. Northern blot analysis revealed that DHC1 is expressed as an approximately 15-kb mRNA in several mammalian cells lines and human tissues, including those that make neither cilia nor flagella.
