Discover sc WTA Kit V2
Discover-scTM WTA Kit V2 is an updated version based on Discover-scTM WTA Kit. The detection sensitivity and volume compatibility have been significantly improved, and it is more suitable for detection of templates of low abundance and low concentration.
Features :
Low starting amount of template : Single cell or 10 pg total RNA can serve as starting templates for efficient amplification.
High sensitivity of amplification : Well optimized reaction system and LNA technology greatly increase the detection rate of low expression genes.
Good integrity of product : F µLl length cDNA is amplified by two end primers, to obtain information of f µLl transcript, avoiding 5'and 3' preferences.
High success rate of operation : Treatment process of RNA sample is greatly reduced, to eliminate the risk of sample loss to the largest extent, and improve the success rate of operation.
Wide compatibility of volume : The compatible sample volume can be as large as 5 μl, compatible with different concentrations of template for amplification.
Low starting amount of template, and good integrity of cDNA amplification
10 pg total RNA of 293T cells was amplified with Discover-scTM WTA Kit V2, and 1 μl cDNA amplification product was analyzed by Agilent 2100 Bioanalyzer. Oligo dT Primer was used as reverse transcription primers, and a linker sequence was added at the 3 'end of cDNA by Template-switching activity of the Discover-sc Reverse Transcriptase. Following PCR amplification employed the linker sequence, to obtain f µLl-length cDNA amplification products, while avoiding the contamination of rRNA.
Library quality analysis - basic data index
Vazyme-1 and Vazyme-2 refer to amplifications of single 293T cell by Discover-scTM WTA Kit V2; C-1 and C-2 refer to amplifications of single 293T cell by single cell transcriptome kit of C company. 1 ng cDNA amplification product were used to construct libraries using TruePrepTM DNA Library Prep Kit V2 for Illumina®. Following information are analyzations of these libraries.

The detection sensitivity of amplification products of Discover-scTM WTA Kit V2 was high, and the number of genes detected in single 293T cell was as high as 15000.
Uniform data distribution without preference

High correlation and repeatability of expression

Discover sc Single Cell Kit V2
After MDA amplification adapted in this kit, the coverage of single cell genome can reach more than 95%, and amplification products Volumes are between 2kb-100kb, with the average product Volume greater than 20 kb, and can be widely used in whole genome and exon sequencing, large copy number variation analysis, microsatellite analysis, qPCR analysis and gene chip analysis.
The Phi 29 DNA polymerase used in this kit is a DNA polymerase cloned from a phage and has strong strand displacement activity, which is capable to achieve melting and replication of DNA chains with the complex structure. In addition, Phi 29 DNA polymerase has strong chain affinity, and one single polymerization process can achieve continuous polymerization and extension of up to 100 kb product. The amplification products are applicable to almost all downstream applications; Phi 29 DNA polymerase also has very strong 3 '-5' exonuclease activity, and its fidelity is 1000 times as that of Taq enzyme, which ensure the high fidelity of DNA synthesis.
Normally, one reaction in Discover-scTM Single Cell and Kit V2 can produce a 30-40 μg genomic DNA of high coverage.

Using qPCR primers distributed on 16 different chromosomes to detect the uniformity and coverage of the whole genome amplification products of Discover-sc Single Cell Kit V2, in which the template was 5 ng. The res µLts showed high uniformity and coverage of the amplified products.

Discover sc Single Cell Kit
The MDA amplification used in this kit to amplify DNA is catalyzed by Phi29 DNA polymerase, which has strong strand displacement activity. This enzyme was cloned from phage and can achieve in vitro thermal-cycle-independent constant temperature DNA polymerization reaction. Single polymerization reaction can achieve up to 100 kb of continuous polymerization. Phi29 DNA Polymerase has strong 3’→5’ exonuclease (proofreading) activity which ensures high fidelity of DNA synthesis, and the fidelity is 1000-fold higher than that of Taq and higher than current most high fidelity polymerases. This kit uses optimized Phi29 DNA polymerase reaction system to achieve constant temperature amplification with high sensitivity of single cell whole genome. This method is suitable for whole genome amplification from single cell, small amount of tissue or minor purified genomic DNA, to obtain massive genomic DNA of high coverage rate.

Discover-sc Single Cell Kit employs optimized reaction system, able to achieve single cell whole genome amplification. When ensuring coverage and uniformity, amplification sensitivity of purified eukaryotic genomic DNA co µLd be 1 ng, and 10 pg for the bacterial genome.
