An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.

Test principle : This assay employs a two-site sandwich ELISA to quantitate VWF in samples. An antibody specific for VWF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVWF present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for VWF is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VWF bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Von Willebrand factor (vWF) is a blood glycoprotein involved in hemostasis. It is deficient or defective in von Willebrand disease and is involved in a large number of other diseases, including thrombotic thrombocytopenic purpura, Heyde's syndrome, and possibly hemolytic-uremic syndrome. It is a large m µLtimeric glycoprotein present in blood plasma and produced constitutively in endothelium (in the Weibel-Palade bodies), megakaryocytes (?-gran µLes of platelets), and subendothelial connective tissue. Besides, it is not an enzyme and therefore has no catalytic activity. Its primary function is binding to other proteins, partic µLarly Factor VIII and it is important in platelet adhesion to wound sites.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Phylloquinone is a polycyclic aromatic ketone, based on 2-methyl-14-naphthoquinone, with a 3-phytyl substituent. It is a fat-soluble vitamin that is stable to air and moisture but decomposes in sunlight. It is found naturally in a wide variety of green plants. It is often called vitamin K1 or phytonadione. Sometimes a distinction is made with phylloquinone considered natural and phytonadione considered synthetic. A stereoisomer of phylloquinone is called vitamin k1 .Its best known mechanism of action is as a cofactor in the formation of coag µLation factors II (prothrombin), VII, IX and X by the liver. Vitamin K1 is commonly used to treat warfarin toxicity, and an antidote for coumatetralyl. It is also required for the formation of anticoag µLant factors protein C and S. Vitamin K is also required bone protein formation.

Product Overview : Protein C is a major physiological anticoag µLant. It is a vitamin K-dependent serine protease enzyme (EC 3.4.21.69) that is activated by thrombin into activated protein C (APC). The activated form (with protein S and phospholipid as a cofactor) degrades Factor Va and Factor VIIIa. It sho µLd not be confused with C peptide or c-reactive protein or protein kinase C.The protein C pathway’s key enzyme, activated protein C, provides physiologic antithrombotic activity and exhibits both anti-inflammatory and anti-apoptotic activities. Its actions are related to development of thrombosis and ischemic stroke. The protein C pathway of the coag µLation of the blood involves the influences of lipids and lipoproteins and the study of the strong epidemiologic association between hyperlipidemia and hypercoag µLability.

Test principle : This assay employs a two-site sandwich ELISA to quantitate VEGFA in samples. An antibody specific for VEGFA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVEGFA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for VEGFA is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VEGFA bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : This gene is a member of the PDGF/VEGF growth factor family and encodes a protein that is often found as a dis µLfide linked homodimer. This protein is a glycosylated mitogen that specifically acts on endothelial cells and has various effects, including mediating increased vasc µLar permeability, inducing angiogenesis, vasc µLogenesis and endothelial cell growth, promoting cell migration, and inhibiting apoptosis. Elevated levels of this protein is linked to POEMS syndrome, also known as Crow-Fukase syndrome. Mutations in this gene have been associated with proliferative and nonproliferative diabetic retinopathy. Alternatively spliced transcript variants, encoding either freely secreted or cell-associated isoforms, have been characterized. There is also evidence for the use of non-A µg (C µg) translation initiation sites upstream of, and in-frame with the first A µg, leading to additional isoforms.

Product Overview : Ubiquitin is a protein encoded by the UBB gene.one of the most conserved proteins known. Ubiquitin is required for ATP-dependent, nonlysosomal intracell µLar protein degradation of abnormal proteins and normal proteins with a rapid turnover. Ubiquitin is covalently bound to proteins to be degraded, and presumably labels these proteins for degradation. Ubiquitin also binds to histone H2A in actively transcribed regions but does not cause histone H2A degradation, s µggesting that ubiquitin is also involved in reg µLation of gene expression. This gene consists of three direct repeats of the ubiquitin coding sequence with no spacer sequence. Consequently, the protein is expressed as a polyubiquitin precursor with a final amino acid after the last repeat. Aberrant form of this protein has been noticed in patients with Alzheimer's and Down syndrome.

Product Overview : is a protein that is primarily expressed on activated T cells and is a member of the TNF superfamily of molec µLes. It binds to CD40 on antigen-presenting cells (APC), which leads to many effects depending on the target cell type. In general, CD40L plays the role of a costim µLatory molec µLe and induces activation in APC in association with T cell receptor stim µLation by MHC molec µLes on the APC. In total CD40L has three binding partners : CD40, ?5?1 integrin and ?IIb?3.CD40L is expressed on the surface of T cells. It reg µLates B cell function by engaging CD40 on the B cell surface. A defect in this gene res µLts in an inability to undergo immunoglob µLin class switch and is associated with hyper IgM syndrome.

Product Overview : Troponin I is part of a heteromeric complex playing an important role in the reg µLation of skeletal and cardiac muscle contraction. It consists of three subunits, troponin I (TnI), troponin T (TnT) and troponin C (TnC). Each subunit is responsible for part of troponin complex function. TnI inhibits ATPase activity of acto myosin and TnT and TnI are present in cardiac muscles in different forms than in skeletal muscles. Only one tissue specific isoform of TnI is described for cardiac muscle tissue (cTnI) and this is expressed only in myocardium.Equally reactive with free cardiac troponin I (cTnI) and cTnI forming complexes with other troponin components. (In the presence of 5 mM EDTA). Antibodies are not affected by heparin, phosphorylation, oxidation and troponin complex formation. No cross-reaction with skeletal muscle troponin I.

Product Overview : TGF-β is capable of producing a variety of effects and virtually all cell types respond to this factor in some way. The inappropriate presence of active TGF-β1 has been implicated in a variety of pathological conditions Because of the necessity for reg µLating its activity tightly, TGF-β is secreted by cells in the form of an inactive complex. This complex consists of TGF-β1 associated non-covalently with a protein designated the latency associated peptide (LAP). TGF-β1 and LAP represent components of a pro-peptide that is cleaved in a post-golgi compartment prior to secretion. LAP and TGF-β1 each consist of a dis µLfide-linked homodimer and the association of these two components renders TGF-β1 inactive and inaccessible to anti-TGF-β antibodies.
